To prepare a 10% dextrose solution with a total volume of 1000 mL follow these steps:

Materials Required:

1.Dextrose (glucose) powder– 100 g  
2.Distilled water– Sufficient quantity to make 1000 mL  
3.Measuring cylinder or volumetric flask (1000 mL)  
4.Balance (to weigh dextrose)  
5.Beaker or container (for mixing)  
6. Stirring rod or magnetic stirrer  
7.pH meter (optional, for adjustment if needed)  
8.Filter (if necessary, for sterilization/clarification)

Procedure:


Step 1: Calculate the Amount of Dextrose Required
- A 10% dextrose solution means 10 g of dextrose in 100 mL of solution.  
- For 1000 mL (1 L) you need:  
 
 

Step 2: Weigh the Dextrose
- Using an analytical balance, accurately weigh 100 g of dextrose powder.

Step 3: Dissolve Dextrose in Water
1. Take about 700–800 mL of distilled water in a beaker or flask.  
2. Gradually add the 100 g dextrose while stirring continuously until fully dissolved.  
3. Ensure no undissolved particles remain.

Step 4: Adjust the Final Volume
- Transfer the solution to a 1000 mL volumetric flask.  
- Rinse the beaker with distilled water and add the rinsings to the flask.  
- Add more distilled water to reach the final volume of 1000 mL.

Step 5: Mix Thoroughly
- Stir well or use a magnetic stirrer to ensure homogeneity.  

Step 6: Sterilization (Optional, if for medical use)
- If needed,filter sterilize using a 0.22 µm membrane filter or autoclave at 121°C for 15 minutes.  

Step 7: Storage
- Store in a clean, airtight container at room temperature or refrigerate if required.  
- Label the solution with:  
  -10% Dextrose Solution 
  - Date of preparation 
  - Expiry date (if applicable)

Final Concentration Verification (Optional)
- Use a refractometer or glucose meter to confirm the 10% concentration.  

Precautions:

✔ Use high-purity dextrose (USP/BP grade) for medical applications.  
✔ Ensure proper dissolution to avoid crystallization.  
✔ If pH adjustment is needed (e.g., for IV use), bring to pH 4–5 with dilute HCl/NaOH (consult pharmacopeia standards).  

This 10% dextrose solution is commonly used for:  
- Intravenous (IV) therapy
- Oral rehydration
- Laboratory experiments
